Toluen Uygulanan Sıçanların Beyin Dokularında Linum usitatissimum, (keten tohumu) Physalis peruviana (altın çilek) ve Ekstraktının Antioksidan Etkileri

Bu çalışmayla in vivo ortamda toluen ile indüklenen doku hasarları üzerine keten tohumu Linum usitatissimum, (Flaxseed) ve altın çilek Physalis peruviana (Goldenberry) örneklerinin antioksidan etkilerinin ortaya çıkarılması ve bu bitkilerin insan sağlığı üzerine olan etkilerinin incelenmesi amaçlanmıştır. Altın çilek ve keten tohumu analizler için ekstrakte edildi. Elde edilen ekstreler toluene uygulanan Sprague Dawley ırkına ait erkek sıçanlara gün aşırı intraperitonal yolla verildikten sonra deney süreleri sonunda dekapite edildi. Beyin dokusu alındıktan sonra oluşturulan lipit fraksiyonunda lipit peroksidasyon (LPO) ölçümü yapıldı. Lipit ekstraktı içindeki yağ asitleri metil esterlerine dönüştürüldükten sonra gaz kromatografisi ile analiz edildi. Ayrıca glutatyon, protein ve vitamin miktarlarıda incelendi. Elde edilen sonuçlara göre; toluen uygulaması sonucu beyin dokularında yağ asidi düzeylerinin yükseldiği saptandı (p<0.01, p<0.001). Toluen uygulamasına karşı bitki ekstraktı verilen gruplarda bütün dokularda glutatyon (GSH) düzeyinin yükseldiği ve malondialdehit (MDA) düzeyinin de azaldığı belirlendi (p<0.05, p<0.01). Beyin dokusunda lipofilik vitamin düzeyleri incelendiğinde, bitki ekstraktı verilen gruplarda α-tokoferol düzeyinin kontrol ve toluen gruplarına göre artmıştır (p<0.05). Deney grubu ile kontrol grubu karşılaştırıldığında beyin dokusundaki kolesterol düzeyi tüm deney gruplarında yükseldi (p<0.05, p<0.001). Verilerimiz keten tohumu ve altın çilek ekstraktının, beyin dokusunda LPO oluşumuna karşı koruyucu etkisi olduğunu gösterdi.

The Antioxidant Effects of Linum usitatissimum and Physalis peruviana Extract in the Brain Tissues of Rats Given Toluene

The purpose of this study is to demonstrate anti-oxidant effects of keten tohumu and altın çilek on toluene-induced tissue damage in vivo and to study the effects of these plant on human health. Physalis peruviana (P) and Flaxseed (F) were extracted for analyses. The extracts obtained were injected intraperitoneally every other day to Spraque male rats given tolüene and the animals were decapitated at the end of the study period. The brain, tissues were removed and lipid peroxidation (LPO) measurements were done in the lipid fraction generated. The fatty acids in the lipid extract were analyzed by gas chromatography after converting them into methyl esters. In addition, the amounts of glutathione, protein, and vitamins were analyzed. Given the results achieved, it was found that the levels of fatty acids increased in the brain tissues after toluene administration (p<0.01, p<0.001). In the groups physalis peruviana and flaxseed against toluene, it was found that the glutathione (GSH) level increased and malondialdehyde (MDA) level reduced in all tissues (p<0.05, p<0.01). Lipophilic vitamin levels examined, plant ekstraxt implement groups levels in the brain tissue increased α-tocopherol compared to control and toluene groups (p<0.05). Experimental groups compared the control group, cholesterol levels in the brain tissues increased all experimental groups. (p<0.05, p<0.001). Our data indicates that keten tohumu ve altın çilek extract has protective effects against LPO formation in the brain tissues.
